The plasmid pSIJ8 is a world-traveller! By now, it has been distributed to over 130 laboratories and companies via Addgene. Thus, the developers of pSIJ8 have been granted the Blue Flame Award.

Researcher Sheila Ingemann Jensen and Professor Alex Toftgaard Nielsen from The Novo Nordisk Foundation Center for Biosustainability at DTU are the happy receivers of a Blue Flame Award from Addgene. The Blue Flame Award serves to congratulate researchers who have at least one plasmid deposited at Addgene that has been distributed more than 100 times.

The reward-winning plasmid, pSIJ8, is based on the so-called Seven-in-Seven method that drastically speeds up the generation of gene deletions in E. coli – for example, up to seven gene deletions can be generated in as little as seven days. With other methods, this number of deletions can take several months to make. Hence, this plasmid enables much faster screening of clones with desired traits, for instance for the production of sustainable bio-products.

Fast generation of deletions in the Seven-in-Seven system is enabled by lambda Red recombineering and a flippase recombinase. Both the plasmid pSIJ8 and the strain SIJ488 are available from Addgene. Currently, 500+ depositors worldwide have one or more Blue Flame plasmids.
